To truly appreciate the area, spend two to three days: base yourself at Glen Helen Resort or camp at Ormiston Gorge, and explore the gorges and walks at a relaxed pace. The drive itself is only about 1.5 hours without stops, but you'll want to linger for walks, swims, and photography. If you're continuing west to Kings Canyon or Uluru, factor in an extra day for the 4WD-only sections beyond Glen Helen.
