Most travellers use Penneshaw as a 1–2 night stopover, arriving by ferry from Cape Jervis and then heading west. Give yourself at least half a day to explore the town's beaches and the penguin centre (book the evening tour in advance). If you're on a self-drive circuit of the island, two nights here allows a relaxed pace to also visit Seal Bay and Flinders Chase National Park further west.
