Drive to Castlemaine and Maldon on day two, enjoying the heritage and short walks. Day three could take you to Bendigo and Daylesford, and if you have a fourth day, head to the Grampians for hiking. The route is around 350 km in total, so you can easily extend with stops at smaller towns like Clunes or Avoca.
